mTOR suppression promotes regulatory T cell development — Delgoffe et al.

Landmark study proving that metabolic state directly determines immune cell identity. Low mTOR = Tregs. High mTOR = effector T cells. The metabolic rationale for caloric restriction in Phase 2.

Effector T cells run on glucose; regulatory T cells run on fat — Michalek et al.

Proved that Tregs and effector T cells use completely different fuel sources. Creating a fat-burning (ketotic) metabolic environment specifically favors Treg development over effector cell production.

Caloric restriction triggers immune system regeneration from stem cells — Cheng et al. (USC)

Caloric restriction does not just optimize existing immune cells — it triggers generation of new immune cells from stem cells, including regulatory T cells. Phase 2 literally rebuilds the immune system.

Ketones directly block inflammatory pathways — Youm et al. (Yale)

Beta-hydroxybutyrate (BHB), the primary ketone body produced in Phase 2, directly blocks the NLRP3 inflammasome — a major driver of inflammatory disease. Ketosis reduces inflammation through a mechanism separate from caloric restriction.

Food changes biology more fundamentally than drugs — Le Couteur et al.

Comparing dietary interventions to pharmaceutical interventions: “Drugs largely dampened responses to diet rather than reshaped them.” Food fundamentally alters protein production. Drugs suppress responses to food. Food is biologically more powerful.
